The 23-time grand slam champion has beaten Czech world number 70 Kristyna Pliskova 7-6 6-4.
It was her first match at one of the 'big four' tournaments since last year's Australian Open after which she gave birth to her first child.
Williams says it was an ace-heavy match so in some ways it was hard to gauge.
She says they got into the rhythm a bit more which certainly helped.
Rafael Nadal has won through in the opening round in straight sets against Italy's Simone Bolelli, as he bids for an 11th men's title.
Defending doubles champion Kiwi Michael Venus has advanced through to the second round with partner Raven Klaasen.
