The fourth seeded Austrian has been bundled out in four sets by Italian Thomas Fabbiano in the first round.
Thiem follows Stefanos Tsitsipas in making an early exit, the 8th seed has lost in four sets to Russian Andrey Rublev.
Spanish 10th seed Roberto Bautista Agut has also crashed out in a day of upsets among the men.
Leading women defending champion Naomi Osaka and fourth seed Simona Halep have survived scares to reach the second round.
Osaka says she struggled to cope with the nerves in her three sets win over Russian Anna Blinkova.
