Defender Kayla Whitelock came out of retirement to play at the Games, but has been ruled out after tearing her anterior cruciate ligament against Australia.
Coach Graham Shaw says they must now reassess the squad they'll take.
Whitelock would have joined Barbara Kendall and potentially Dame Valerie Adams as the only Kiwi women to attend five Olympics.
The men's and women's squads are named next week.
